摘要:
目的:对人食管鳞癌细胞及正常人食管上皮细胞进行代谢组学分析,筛选特征代谢标志物。方法:采用基于气相色谱-质谱联用的代谢组学方法,对3株食管鳞癌细胞EC1、Eca109、TE1及正常食管上皮细胞Het-1A中小分子代谢产物进行检测,利用无监督的主成分分析及单因素方差分析,根据组间代谢群组的代谢途径及代谢物的差异探讨食管鳞癌细胞的代谢特征。结果:EC1、Eca109、TE1和Het-1A相比,代谢模式存在明显差异,差异代谢物有赖氨酸、酪氨酸、缬氨酸、蛋氨酸、谷氨酸、葡萄糖、塔格糖及胆固醇等,分别涉及氨基酸代谢、糖代谢和能量代谢等多个代谢途径。结论:通过代谢组学研究筛选出了食管鳞癌细胞中的差异代谢物。
Abstract:
Aim:Metabolomic analysis of human esophageal squamous cell carcinoma and normal esophageal epithelial cells was carried out to screen for characteristic metabolic markers.Methods:GC-MS was used to identify the differential metabolites between esophageal squamous cell carcinoma cell lines EC1, Eca109, TE1 and normal esophageal epithelial cell Het-1A. Unsupervised principal component analysis and one-way ANOVA were used to analyze the differential metabolites, and the metabolic characteristics of esophageal squamous cell carcinoma cells based on the differences of metabolic pathways and metabolites between cells were discussed.Results:The metabolites of EC1, Eca109 and TE1 showed significantly difference compared with Het-1A, including lysine, tyrosine, valine, methionine, glutamic acid, glucose, tagatose and cholesterol, which were involved in amino acid metabolism, glucose metabolism and energy metabolism.Conclusion:Application of metabolomics successfully found differential metabolites of esophageal squamous cell carcinoma cells.
