摘要:
目的:构建胃腺癌竞争性内源RNA(ceRNA)网络,并观察miR-204-5p和HOXC8 mRNA之间的相关性。方法:利用生物信息学方法和TCGA转录组数据库筛选胃腺癌差异基因,构建ceRNA网络,检测了miR-204-5p和HOXC8 mRNA在胃腺癌组织和SGC7901细胞株中的表达,并分析了两基因间的相互关系。结果:成功构建胃腺癌ceRNA网络,miR-204-5p是重要节点基因,并预测到其靶基因HOXC8; 在32例胃腺癌中检测到miR-204-5p低表达,HOXC8高表达,两者呈负相关关系; 上调胃腺癌SGC7901细胞中miR-204-5p的表达可以抑制HOXC8 mRNA的表达。结论:miR-204-5p和HOXC8可能是潜在的胃腺癌诊断指标,两者可能存在ceRNA调控关系。
Abstract:
Aim:To construct the competing endogenous RNA(ceRNA)network of gastric adenocarcinoma(GAC)and observe the correlation between miR-205-5p and HOXC8 mRNA in GAC.Methods:The ceRNA network of GAC was constructed by using bioinformatics method based on the data of transcriptome of GAC in TCGA database. The expression levels of miR-204-5p and HOXC8 mRNA were detected by real-time PCR in tissue specimens of GAC and SGC7901 cells. The correlation between the two genes was analyzed.Results:The ceRNA network of GAC was successfully constructed. MiR-204-5p was an important node gene, and its target gene HOXC8 was found from the network. Low expression of miR-204-5p and high expression of HOXC8 were detected in 32 GAC tissue samples,and there was a negative correlation between them. MiR-204-5p mimics transfection cells could significantly inhibited the expression of HOXC8 mRNA in SGC7901 cells.Conclusion:miR-204-5p and HOXC8 might be potential diagnostic markers of GAC, and the ceRNA network composed of two genes might be exist in GAC.
