摘要:
目的:探讨COX-2抑制剂对前列腺炎(CP)大鼠射精时间的影响及其机制。方法:雄性Wistar大鼠随机分为模型组(n=12)、正常对照组(n=10)和COX-2抑制剂组(n=10)。模型组和COX-2抑制剂组大鼠自身免疫法建立CP模型。造模成功后,模型组大鼠18 mg/(kg·d)灌胃塞来昔布,正常对照组和COX-2抑制剂组给予生理盐水。连用4周后,进行交配实验,记录骑跨潜伏期(ML)、插入潜伏期(IL)及射精潜伏期(EL)。各组大鼠行前列腺组织HE染色; ELISA法检测血清TNF-α、IL-1β水平; 免疫组化和Western blot法检测T13~L2和L5~S2段脊髓5-羟色胺(5-HT)、5-HT1A、5-HT2C及5-HT转运(5-HTT)蛋白的表达。结果:模型组大鼠表现出典型的前列腺炎症。与正常对照组比较,模型组大鼠 EL缩短,血清IL-1β和TNF-α升高,T13~L2及L5~S2脊髓5-HT水平下降、5-HT1A水平升高,L5~S2脊髓5-HTT升高(P<0.05)。而COX-2抑制剂组前列腺炎症轻微,血清TNF-α、IL-1β水平、脊髓5-HT及其受体水平、EL均较模型组改善(P<0.05)。结论:CP大鼠EL缩短与前列腺炎症有关。COX-2抑制剂可以延长CP大鼠EL。
Abstract:
Aim:o investigate the effect of COX-2 inhibitor on ejaculation time of rats with chronic prostatitis(CP)and its mechanism.Methods:Male Wistar rats were randomly divided into model group(n=12), normal control group(n=10), and COX-2 inhibitor group(n=10). CP rat models were established by autoimmune method. After 4 weeks for modeling, 18 mg/(kg·d)celecoxib were administered by gavage in the model group, and physiological saline were used in the normal control and COX-2 inhibitor groups. After that, a mating experiment was performed to record the riding latency(ML), insertion latency(IL), and ejaculation latency(EL). HE of prostate tissue was performed. Serum TNF-α and IL-1β were detected by ELISA. Immunohistochemistry and Western blot were used to detect 5-hydroxytryptamine(5-HT), 5-HT1A, 5-HT2C and 5-HT transporter(5-HTT)in the spinal cord of T13~L2 and L5~S2.Results:Prostatic inflammation were showed in the model group. Compared with the normal group, EL was shortened, serum IL-1β and TNF-α were increased, and 5-HT levels in T13~L2 and L5~S2 spinal cord were decreased. The level of 5-HT1A was elevated, and the 5-HTT of L5~S2 spinal cord was elevated(P<0.05). In the COX-2 inhibitor group, prostate inflammation was mild, cytokine levels, spinal cord 5-HT and its receptor levels, and EL were improved compared with the normal control group(P<0.05).Conclusion:EL shortening in CP rats is associated with prostatic inflammation and pelvic pain. COX-2 inhibitors can prolong rat EL.
