摘要:
目的:探讨上调睾丸特异性Y样蛋白5(TSPYL5)基因的表达对结直肠癌细胞侵袭、迁移及STAT3蛋白表达的影响。方法:将人结直肠癌LoVo细胞分为空白1组、空载体组和TSPYL5质粒组(转染pcDNA3.1-TSPYL5质粒)。转染48 h后,采用Transwell小室检测细胞侵袭和迁移能力,采用Western blot法检测TSPYL5、E-cadherin、Vimentin、STAT3和p-STAT3蛋白的表达。另取LoVo细胞分为空白2组、AG490(STAT3信号抑制剂)组和AG490+TSPYL5质粒组。转染48 h后,采用Transwell小室检测细胞侵袭和迁移能力,采用Western blot法检测E-cadherin和Vimentin蛋白的表达。结果:与空白1组和空载体组相比,TSPYL5质粒组细胞侵袭及迁移能力降低,TSPYL5蛋白表达水平升高,E-cadherin蛋白表达水平升高,Vimentin及p-STAT3蛋白表达水平降低(P均<0.05)。与空白2组相比,AG490组细胞侵袭和迁移能力、Vimentin蛋白表达水平均降低,E-cadherin蛋白的表达水平升高; 与AG490组相比,AG490+TSPYL5质粒组细胞侵袭、迁移能力及Vimentin蛋白的表达水平进一步降低,E-cadherin蛋白的表达水平升高(P均<0.05)。结论:上调TSPYL5的表达可能抑制STAT3蛋白的表达,逆转结直肠癌细胞EMT,从而降低细胞侵袭和迁移能力。
Abstract:
Aim:To investigate the effects of up-regulation of TSPYL5 on invasion, migration and STAT3 protein in colorectal cancer cells.Methods:Human colorectal cancer LoVo cells were divided into blank group 1, empty vector group and TSPYL5 group(transfected with pcDNA3.1-TSPYL5 plasmid). After 48 hours, the cell invasion and migration were detected by Transwell chamber,and the expressions of TSPYL5, E-cadherin, Vimentin, STAT3 and p-STAT3 proteins were detected by Western blot. LoVo cells were divided into blank group 2, AG490(STAT3 signaling pathway inhibitor)group and AG490+TSPYL5 group. Transwell chamber was used to detect the invasion and migration ability. Western blot was used to detect the expressions of E-cadherin and Vimentin proteins.Results:Compared with blank group 1 and empty vector group, the expression of TSPYL5 in TSPYL5 group was higher, the cell invasion and migration ability decreased, the expression of E-cadherin increased, while those of Vimentin and p-STAT3 decreased(P<0.05). Compared with blank group 2,the invasion and migration ability of cells in AG490 group decreased, the expression of Vimentin was lower while that of E-cadherin was higher(P<0.05); compared with AG490 group, the invasion and migration ability and the expression of Vimentin in AG490+ TSPYL5 group decreased while the expression of E-cadherin increased(P<0.05).Conclusion:Up-regulation of TSPYL5 can inhibit the expression of STAT3,reverse epithelial mesenchymal transition in colorectal cancer cells, and thereby reduce invasion and migration ability of cells.
