摘要:
目的:探讨沉默鼠双微体4(MDM4)基因对卵巢癌细胞顺铂(DDP)耐药性的影响。方法:采用qRT-PCR法检测卵巢癌细胞株SKOV3、A2780及其DDP耐药细胞株SKOV3/DDP、A2780/DDP中MDM4的表达情况。在A2780/DDP细胞中分别转染MDM4 siRNA及阴性对照,分别记为MDM4 siRNA组和阴性对照组,将未转染的A2780/DDP细胞记为空白对照组,采用qRT-PCR和Western blot法检测转染效率,采用MTT法检测细胞增殖抑制率,采用Western blot法检测t-PI3K、p-PI3K、t-AKT和p-AKT蛋白的表达水平。结果:SKOV3/DDP、A2780/DDP细胞与SKOV3、A2780细胞比较,MDM4的表达水平升高(P<0.05)。MDM4 siRNA组与空白对照组和阴性对照组相比,MDM4 mRNA和蛋白表达水平降低,细胞增殖抑制率升高,p-PI3K和p-AKT蛋白表达水平降低,IC50降低(P<0.05)。结论:沉默MDM4基因能够降低A2780/DDP细胞对DDP的耐药性,其作用机制可能与抑制PI3K/AKT信号通路的激活有关。
Abstract:
Aim:To investigate the effects of MDM4 gene on cisplatin resistance in ovarian cancer cells.Methods:The expression of MDM4 in ovarian cancer cell lines SKOV3, A2780 and ovarian cancer cisplatin-resistant cell lines SKOV3/DDP and A2780/DDP was detected by qRT-PCR. MDM4 siRNA and negative control were transfected into A2780/DDP cells, respectively, and recorded as MDM4 siRNA group and negative control group, and untransfected A2780/DDP cells were recorded as blank control group. The transfection efficiency was detected by qRT-PCR and Western blot. The proliferation inhibition rate of A2780/DDP cells was detected by MTT assay. Western blot was used to detect the expression levels of t-PI3K, p-PI3K, t-AKT and p-AKT proteins.Results:Compared with ovarian cancer cells SKOV3 and A2780, the expression of MDM4 was significantly increased in ovarian cancer cisplatin-resistant cells SKOV3/DDP and A2780/DDP(P<0.05). Compared with blank control group and negative control group, the MDM4 mRNA and protein expressions were significantly decreased in MDM4 siRNA group, cell proliferation inhibition rate was significantly increased,p-PI3K and p-AKT protein expression levels were significantly decreased,and IC50 decreased(P<0.05).Conclusion:Silencing MDM4 gene could reduce the resistance of A2780/DDP cells to cisplatin, and its mechanism may be related to the inhibition of PI3K/AKT signaling pathway activation.
