摘要:
目的:研究miR-130a对缺氧诱导的人肺动脉平滑肌细胞(HPASMC)增殖的影响。方法:采用qRT-PCR检测合并肺动脉高压和肺动脉压正常的先天性心脏病患儿血清以及在常氧和缺氧培养条件下HPASMC中miR-130a的表达水平。对缺氧诱导的HPASMC转染miR-130a抑制剂(miR-130a抑制物组)或抑制剂对照(阴性对照组),未转染细胞作为空白对照组,采用qRT-PCR法检测转染效率,采用EdU法检测细胞增殖情况,采用Western blot法检测细胞中Wnt1和β-catenin蛋白表达水平。结果:与肺动脉压正常的先天性心脏病患儿比较,先天性心脏病合并肺动脉高压患儿血清中miR-130a水平升高(P<0.001)。与常氧组相比,缺氧组HPASMC中miR-130a表达水平升高(P<0.001)。与空白对照组、阴性对照组比较,miR-130a 抑制物组HPASMC中miR-130a表达水平降低,细胞增殖能力降低,Wnt1和β-catenin蛋白表达水平上调(P<0.05)。结论:miR-130a能够抑制缺氧诱导的HPASMC增殖,其作用机制可能与Wnt信号通路的激活有关。
Abstract:
Aim:To investigate the effects of miR-130a on hypoxia-induced proliferation of human pulmonary artery smooth muscle cell(HPASMC).Methods:qRT-PCR was used to detect the serum level of miR-130a in patients with congenital heart disease(CHD)and pulmonary hypertension and CHD patients with normal pulmonary artery pressure and in HPASMC under normoxia and hypoxia. HPASMC was divided into miR-130a inhibitor group,negative control group,and blank control group. The transfection efficiency was detected by qRT-PCR. Cell proliferation was analyzed by EdU method. Wnt1 and β-catenin protein expression levels were detected by Western blot.Results:Compared with normal pulmonary artery pressure CHD patients, the expression of miR-130a in serum of patients with CHD and pulmonary hypertension was significantly increased(P<0.001). Compared with normoxia, the expression of miR-130a in hypoxia-induced HPASMC was significantly increased(P<0.001). Compared with the other two groups, the expression level of miR-130a was significantly decreased in the miR-130a inhibitor group, the cell proliferation ability was significantly decreased, and the expression levels of Wnt1 and β-catenin were significantly up-regulated(P<0.05).Conclusion:miR-130a could inhibit hypoxia-induced proliferation of HPASMC, and its mechanism may be related to the activation of Wnt signaling pathway.
