摘要:
目的:采用脂质组学方法研究哮喘小鼠肺组织中甘油酯类代谢的差异性改变。方法:将16只6周龄无特定病原体级BALB/c 雄性小鼠分为哮喘组和对照组,每组8只。哮喘组小鼠给予卵清蛋白联合氢氧化铝诱导哮喘模型,对照组采用PBS替代。造模结束24 h后采用无创法检测乙酰甲胆碱激发后小鼠的增强呼气间歇(Penh)值以评估气道反应性。收集小鼠支气管肺泡灌洗液(BALF)进行细胞分类计数,检测IL-4、IL-5、IL-13、胸腺基质淋巴细胞生成素的水平。通过液相色谱/质谱联用技术检测小鼠肺组织中甘油酯类成分。结果:哮喘组小鼠的Penh值高于对照组(P=0.011)。哮喘组小鼠BALF中炎症细胞总数、嗜酸性细胞计数以及IL-4、IL-5、IL-13、胸腺基质淋巴细胞生成素水平均较对照组升高(P<0.05)。与对照组小鼠比较,哮喘组小鼠肺组织中有12种甘油酯类表达升高(P<0.05)。结论:卵清蛋白诱导的BALB/c哮喘小鼠肺组织中存在甘油酯类代谢紊乱。
Abstract:
Aim:To study the glycerides metabolism difference in the lung tissue of asthmatic mice by using lipidomics.Methods:Sixteen 6-week specific pathogen-free grade BALB/c male mice were randomly allocated into asthma group and control group,with 8 mice in each group.Mice in the asthma group were induced by ovalbumin combined with aluminum hydroxide to build asthma model,and the mice in control group were replaced with PBS.After 24 hours of the modeling,the enhanced pause(Penh)value of the mice after methacholine challenging was measured by non-invasive method to assess airway hyperresponsiveness.Mouse bronchoalveolar lavage fluid was collected for cell counts,and the levels of IL-4,IL-5,IL-13 and thymic stromal lymphopoietin were detected.The glyceride species in mice lung tissue were detected by liquid chromatography-mass spectrometry.Results:The Penh values of the asthma group were higher than those of the control group(P=0.011).The total number of inflammatory cells,eosinophil count and the levels of IL-4,IL-5,IL-13,thymic stromal lymphopoietin in bronchoalveolar lavage fluid of mice in the asthma group were higher than those of control group(P<0.05).The expression levels of 12 glycerides were increased in the lung tissue of the asthma group compared with the control group(P<0.05).Conclusion:There is glycerides metabolism disorder in the lung tissue of ovalbumin-induced BALB/c asthmatic mice.
