摘要:
目的:探讨LncRNA IRAIN过表达对急性髓系白血病(AML)细胞增殖及凋亡的影响。方法:收集人AML细胞系HL-60、NB4、U937、K562、OCI-AML2、OCI-AML5、Kasumi-6和原代正常骨髓单个核细胞(BMMC),实时荧光定量PCR法检测细胞中IRAIN的表达。利用Lipofectamine3000将IRAIN过表达载体转染入NB4细胞(IRAIN过表达组),以未转染细胞为空白对照组,转染无序对照物的细胞为阴性对照(NC)组,实时荧光定量PCR法检测3组细胞中IRAIN的表达,分别采用MTT法、流式细胞术和Annexin V染色法检测3组细胞增殖活性、细胞周期及凋亡率,Western blot法检测细胞中IGF1R、IGF1、IGF2、Akt蛋白的表达。结果:与正常BMMC比较,HL-60、NB4、OCI-AML2、OCI-AML5及Kasumi-6细胞中IRAIN表达下调,NB4细胞中表达水平最低(P<0.05)。与空白对照组和NC组比较,IRAIN过表达组细胞中IRAIN表达水平升高(P<0.05); 细胞增殖活性降低,凋亡率增加,细胞周期被阻滞于G0/G1期(P<0.05); 细胞中IGF1、IGF2蛋白表达水平降低,IGF1R、Akt磷酸化水平增加(P<0.05)。结论:LncRNA IRAIN可能通过影响IGF1R转录及表达,干扰AML细胞的增殖及凋亡。
Abstract:
Aim:To investigate the effects of LncRNA IRAIN overexpression on the proliferation and apoptosis of acute myeloid leukemia(AML)cells.Methods:The expression of IRAIN in HL-60,NB4,U937,K562,OCI-AML2,OCI-AML5,Kasumi-6 and normal BMMC was detected by real-time quantitative PCR.The NB4 cells in IRAIN overexpression group were transfected with IRAIN overexpression vector by Lipofectamine3000.The cells without transfection were the blank control,and the cells transfected with disordered vector were the negative control(NC).The expression of IRAIN in the cells of the 3 groups was detected by real-time quantitative PCR; cell proliferation,cell cycle and apoptosis rate were measured by MTT assay,flow cytometry and Annexin V staining; and the expressions of IGF1R,IGF1,IGF2,Akt proteins were detected by Western blot.Results:Compared with the normal BMMC,the expression of IRAIN in HL-60,NB4,OCI-AML2,OCI-AML5 and Kasumi-6 cells decreased,which was the lowest in NB4 cells(P<0.05).Compared with the blank control group and NC group,the expression of IRAIN in the IRAIN overexpression group increased(P<0.05); the cell proliferation activity decreased,the apoptosis rate increased,and the cell cycle was arrested in G0/G1 phase(P<0.05); the expressions of IGF1 and IGF2 proteins were lower,and those of IGF1R and Akt phosphorylation increased(P<0.05).Conclusion:LncRNA IRAIN may interfere with the proliferation and apoptosis of AML cells by affecting the transcription and expression of IGF1R.
