摘要:
目的:分析慢性阻塞性肺疾病(COPD)急性加重期患者血清分泌型卷曲相关蛋白1(SFRP1)水平的变化及意义。方法:COPD患者118例,其中急性加重期43例,稳定期75例; 同期体检健康成年人50例(正常对照组)。ELISA法检测研究对象血清SFRP1和炎症反应相关指标IL-17、IL-33、脂联素(APN)水平,放射免疫法检测气道重塑相关指标(MMP-9)、基质金属蛋白酶抑制因子-1(TIMP-1)、神经生长因子(NGF)、碱性成纤维细胞生长因子(b-FGF)、血管内皮生长因子(VEGF)水平,分析血清SFRP1水平与炎症反应相关指标、气道重塑相关指标的相关性。结果:COPD急性加重期组和稳定期组血清SFRP1水平、IL-17和IL-33水平,以及5种气道重塑相关指标均高于正常对照组,APN水平低于正常对照组(P<0.05); COPD急性加重期组上述指标的变化大于稳定期组(P<0.05)。Pearson相关分析结果显示,COPD患者血清SFRP1水平与APN呈负相关(r=-0.426,P<0.05); 与IL-17、IL-33及MMP-9、TIMP-1、VEGF、NGF、b-FGF水平均呈正相关(r=0.306、0.315、0.395、0.328、0.305、0.462、0.506,P<0.05)。结论:COPD患者血清SFRP1水平升高,并与病情急性加重、炎症反应激活、气道重塑加剧有关。
Abstract:
Aim:To analyze the changes of serum secreted frizzled-associated protein 1(SFRP1)level in patients with acute exacerbation chronic obstructive pulmonary disease(COPD)and its clinical significance.Methods:A total of 118 COPD patients(43 cases in acute exacerbation stage and 75 cases in stable stage)and 50 healthy adults(normal control group)were subjected to detect the serum levels of SFRP1 and indicators related to inflammatory response(IL-17,IL-33,adiponectin)by ELISA,the serum levels of airway remodeling related indicators including MMP-9,vascular endothelial growth factor(VEGF),matrix metalloproteinase inhibitor factor-1(TIMP-1),nerve growth factor(NGF),and basic fibroblast growth factor(b-FGF)by radioimmunoassay.The correlations between SFRP1 level and those of indicators related to inflammatory response and indicators related to airway remodeling were assessed using Pearson relation analysis.Results:The levels of SFRP1,IL-17,IL-33,MMP-9,TIMP-1,VEGF,NGF,b-FGF in the acute exacerbation and stable groups were significantly higher than those in the normal control group(P<0.05),while the level of adiponectin was significantly lower than that in the normal control group(P<0.05).The changes of the indicators mentioned above of the acute exacerbation group were much more than the stable group(P<0.05).The correlation analysis showed that the serum SFRP1 content in patients with COPD was negatively correlated with adiponectin(r=-0.426,P<0.05),positively related to IL-17,IL-33 and MMP-9,TIMP-1,VEGF,NGF,b-FGF(r=0.306,0.315,0.395,0.328,0.305,0.462,0.506,P<0.05).Conclusion:The serum SFRP1 level of COPD patients is significantly higher,which is related to the acute exacerbation,activation of inflammatory response,and exacerbation of airway remodeling.
