摘要:
目的:探讨类风湿关节炎(RA)外周血单个核细胞(PBMC)中微小RNA-142-3p(miR-142-3p)的表达对RA成纤维样滑膜细胞(FLS)增殖和凋亡的影响。方法:采用生物信息学预测和双荧光素酶报告实验分析miR-142-3p和解聚素金属蛋白酶17(ADAM17)的靶向关系。分别采用实时荧光定量PCR法和Western blot法检测人正常PBMC和RA-PBMC中miR-142-3p和ADAM17 蛋白的表达。利用miR-142-3p模拟物或抑制物转染调控RA-PBMC中miR-142-3p的表达,观察低表达miR-142-3p,及过表达ADAM17和miR-142-3p的RA-PBMC对RA-FLS增殖和凋亡的影响。RA-FLS增殖的检测采用MTT法,RA-FLS凋亡的检测采用Annexin V-FITC和PI双染法。结果:ADAM17和miR-142-3p存在靶向关系。与正常PBMC相比, RA-PBMC中miR-142-3p表达上调,ADAM17蛋白表达下调(P<0.05)。低表达miR-142-3p的RA-PBMC和过表达ADAM17的RA-PBMC均可增强RA-FLS增殖活性,减少其凋亡(P<0.05)。过表达miR-142-3p的RA-PBMC可降低RA-FLS的增殖活性,促进其凋亡(P<0.05)。过表达ADAM17 和过表达miR-142-3p的作用可以相互拮抗(P<0.05)。结论:调控RA-PBMC中miR-142-3p的表达可以通过调控ADAM17而影响RA-FLS的增殖和凋亡。
Abstract:
Aim:To investigate the effects of miR-142-3p expression in peripheral blood mononuclear cells(PBMC)from the patients with rheumatoid arthritis(RA)on the proliferation and apoptosis of fibroblast-like synovial cells(FLS)of RA patients.Methods:The targeting relationship between miR-142-3p and metalloproteinase 17(ADAM17)was analyzed by biological information prediction and double luciferase report test.The expressions of miR-142-3p and ADAM17 in normal PBMC and RA-PBMC were detected by real-time quantitative PCR and Western blot,respectively.miR-142-3p expression in RA-PBMC was regulated by miR-142-3p analogue or inhibitor transfection.The effects of RA-PBMC with low miR-142-3p expression,or overexpression of ADAM17 and miR-142-3p on proliferation and apoptosis of RA-FLS were observed.The proliferation of RA-FLS was detected by MTT assay,and the apoptosis of RA-FLS was detected by Annexin V-FITC and PI double staining.Results:There was a targeted relationship between ADAM17 and MIR-142-3P.Compared with normal PBMC,the expression of miR-142-3p in RA-PBMC was higher,and that of ADAM17 was lower(P<0.05).The proliferation of RA-FLS induced by RA-PBMC with low expression of miR-142-3p or overexpression of ADAM17 decreased and the apoptosis rate increased(P<0.05).The proliferation of RA-FLS induced by RA-PBMC with overexpression of miR-142-3p increased,the apoptosis rate decreased(P<0.05).Overexpression of ADAM17 and overexpression of miR-142-3p could antagonize each other(P<0.05).Conclusion:Modulating the expression of miR-142-3p in RA-PBMC could affect the proliferation and apoptosis of RA-FLS through modulating ADAM17.
