摘要:
目的:研究虫草素对脂多糖(LPS)处理的鼻黏膜上皮细胞增殖和细胞因子表达的影响。方法:分离培养人鼻黏膜上皮细胞,用0、1、2、4、8 mg/L的虫草素处理24 h,采用MTT法检测细胞增殖活性。将细胞分为对照组(不处理)、LPS组(100 mg/L LPS处理48 h)和LPS+虫草素组(100 mg/L LPS处理48 h后加入4 mg/L虫草素作用24 h),采用MTT法检测细胞增殖活性,qRT-PCR法检测细胞中IL-1β、TNF-α、ICAM-1 mRNA的表达,Western blot法检测细胞中NF-κB信号通路关键蛋白p-IκBα、p-P65蛋白的表达。结果:虫草素可促进鼻黏膜上皮细胞增殖,4 mg/L虫草素对鼻黏膜上皮细胞的增殖促进作用最明显。与对照组相比,LPS组细胞增殖活性下降,IL-1β、TNF-α、ICAM-1 mRNA和p-IκBα、p-P65蛋白的表达升高(P<0.05); 与LPS组相比,LPS+虫草素组细胞增殖活性升高,IL-1β、TNF-α、ICAM-1 mRNA和p-IκBα、p-P65蛋白的表达降低(P<0.05)。结论:虫草素可以促进鼻黏膜上皮细胞增殖,可能与抑制NF-κB信号通路和下游细胞因子的表达有关。
Abstract:
Aim:To study the effect of cordycepin on the cell proliferation and expression of cytokines in LPS-induced nasal mucosa epithelial cells.Methods:The effect of cordycepin at different doses(0,1,2,4,8 mg/L)on the proliferation of human nasal mucosa epithelial cells was detected by MTT method.The cells were divided into three groups,control group(not treated),LPS group(treated with 100 mg/L LPS for 48 hours),and LPS+cordycepin group(4 mg/L cordycepin for 24 hours after being treated with 100 mg/L LPS for 48 hours).The proliferation of nasal mucosa epithelial cells was detected by MTT method.The expressions of IL-1β,TNF-α and ICAM-1 mRNA in the cells were detected by qRT-PCR.The expressions of p-IκBα and p-P65,the key proteins of NF-κB signaling pathway,were detected by Western blot.Results:The proliferation activity of nasal mucosa epithelial cells increased after cordycepin treatment,especially the cordycepin at 4 mg /L.Compared with the control group,the proliferation activity of LPS group was decreased,the expressions of IL-1β,TNF-α,ICAM-1 mRNA and p-IκBα,p-P65 protein were increased significantly(P<0.05).Compared with LPS group,the proliferation activity of LPS+cordycepin group was increased,the expressions of IL-1β,TNF-α,ICAM-1 mRNA and p-IκBα,p-P65 protein were decreased(P<0.05).Conclusion:Cordycepin can promote the proliferation of LPS induced nasal mucosa epithelial cells.The possible mechanism is that cordycepin can inhibit the expression of cytokines in downstream by inhibiting the NF-κB signaling pathway.
