摘要:
目的:研究丹酚酸B对金黄色葡萄球菌肺炎幼年小鼠(幼鼠)肺组织修复及脑神经保护的作用及机制。方法:选择125只幼鼠,除25只幼鼠作为对照组外,其余100只幼鼠采用鼻滴金黄色葡萄球菌菌液的方法建立金黄色葡萄球菌肺炎幼鼠模型,造模完成后的幼鼠分为模型组和丹酚酸B低、中、高剂量组,每组25只,丹酚酸B低、中、高剂量组幼鼠分别按照5、10、20 mg/kg腹腔注射丹酚酸B,对照组及模型组腹腔注射等体积生理盐水,1次/d,连续7 d。采用Morris水迷宫实验测定幼鼠学习记忆能力,使用ELISA试剂盒检测幼鼠血清TNF-α、IL-6、IL-1β水平,血气分析仪检测动脉氧分压(PaO2)、氧合指数(OI),计算肺组织湿干重比值,化学显色法测定肺组织SOD活性及MDA水平,HE染色后观察幼鼠肺组织病理学变化,Western blot法检测肺组织中NF-κBp65、p-NF-κBp65蛋白表达水平,免疫荧光法检测脑组织小胶质细胞标记物离子钙接头分子(Iba-1)蛋白表达水平。结果:与对照组相比,模型组幼鼠逃避潜伏期及首次穿过平台时间延长,90 s内穿过平台次数减少; 血清TNF-α、IL-6、IL-1β水平,肺组织湿干重比值,肺组织MDA水平,肺组织NF-κBp65、p-NF-κBp65蛋白表达水平及p-NF-κBp65/NF-κBp65比值,脑组织Iba-1蛋白表达水平升高; PaO2、OI、肺组织SOD活性降低(P<0.05)。与模型组相比,丹酚酸B低、中、高剂量组幼鼠逃避潜伏期及首次穿过平台时间缩短,90 s内穿过平台次数增加; 血清TNF-α、IL-6、IL-1β水平,肺组织湿干重比值,肺组织MDA水平,肺组织NF-κBp65、p-NF-κBp65蛋白表达水平及p-NF-κBp65/NF-κBp65比值,脑组织Iba-1蛋白表达水平降低; PaO2、OI、肺组织SOD活性升高(P<0.05)。结论:丹酚酸B能够减轻金黄色葡萄球菌肺炎幼鼠体内炎症状态,改善肺组织及脑组织炎症水平,保护脑神经,提高学习记忆能力。
Abstract:
Aim:To study the effects and mechanism of Salvianolic acid B on lung tissue repair and cerebral nerve protection in mouse pups with Staphylococcus aureus pneumonia.Methods:Except for 25 mouse pups as control group, the other 100 mouse pups were used to establish the model of Staphylococcus aureus pneumonia by nasal dripping Staphylococcus aureus,and then randomly allocated into model group and low, middle,high dose Salvianolic acid B groups(n=25).The mouse pups in low, middle and high dose groups were intraperitoneally injected with Salvianolic acid B at 5, 10 and 20 mg/kg, respectively,and the control group and the model group were given normal saline,once a day for 7 days.The learning and memory abilities were measured by Morris water maze test.The levels of serum TNF-α, IL-6 and IL-1β were detected by ELISA kit.The partial pressure of arterial oxygen(PaO2)and oxygenation index(OI)were measured by blood gas analyzer.The wet/dry weight ratio of lung tissue was calculated.The activity of SOD and the level of MDA in lung tissue were measured by chemical colorimetric method.The pathological changes of lung tissue were observed after HE staining.The protein expression levels of NF-κBp65 and p-NF-κBp65 in lung tissue were detected by Western blot.The expression level of microglial marker ion calcium junction molecule(Iba-1)in brain tissue was detected by immunofluorescence.Results:Compared with the control group, the escape latency and the time of crossing the platform for the first time in the model group were prolonged, and the times of crossing the platform within 90 seconds were decreased(P<0.05).The levels of serum TNF-α, IL-6, IL-1β, wet/dry weight ratio of lung tissue, MDA level in lung tissue, the expression levels of NF-κBp65, p-NF-κBp65 protein in lung tissue and the ratio of p-NF-κBp65/NF-κBp65, the expression level of Iba-1 protein in brain tissue increased; while PaO2, OI and the activity of SOD in lung tissue decreased(P<0.05).Compared with the model group, the escape latency and the time of crossing the platform for the first time in the low, middle and high dose groups of Salvianolic acid B were shortened, and the times of crossing the platform within 90 seconds were increased(P<0.05); the levels of serum TNF-α, IL-6, IL-1β, wet/dry weight ratio of lung tissue, MDA level in lung tissue, the expression levels of NF-κBp65 and p-NF-κBp65 protein in lung tissue, the ratio of p-NF-κBp65/NF-κBp65, and Iba-1 level in brain tissue decreased in Salvianolic acid B low, middle and high dose groups(P<0.05); the PaO2, OI and the activity of SOD in lung tissue increased(P<0.05).Conclusion: Salvianolic acid B could reduce the inflammatory state in mouse pups with Staphylococcus aureus pneumonia, improve the inflammatory level of lung and brain tissue, protect the brain nerves and improve the learning and memory ability.
