摘要:
目的:观察顺铂(cDDP)对肿瘤坏死因子相关凋亡诱导配体(TRAIL)诱导EC1细胞凋亡的增敏效应。方法:MTT法检测单用TRAIL(10、50、100、500和1 000 μg/L)或cDDP(1.0、2.0、4.0、10.0和20.0 mg/L)分别作用24、48和72 h 对EC1细胞增殖的影响。流式细胞术检测100 μg/L TRAIL和10.0 mg/L cDDP单用或联用48 h对EC1细胞凋亡和细胞周期的影响,并设空白对照组。结果:单用TRAIL作用24、48和72 h对EC1细胞增殖影响不明显(P>0.05),单用cDDP对EC1细胞增殖有抑制作用(P<0.05)。单用TRAIL对EC1细胞凋亡率无影响(FTRAIL=5.836,P=0.250),而单用cDDP可提高EC1细胞凋亡率(FcDDP=20.763,P=0.041),二者联合有交互作用(F交互=57.422,P<0.001)。单用TRAIL和cDDP对EC1细胞周期无影响,二者联用后G0/G1和S期细胞比例下降,G2/M细胞比例上升(P均<0.001)。结论:cDDP可增加EC1细胞对TRAIL的敏感性,二者联用可诱导EC1细胞凋亡,并诱导其在G2/M期阻滞。
